鸡的大小黄白卵泡分离及总RNA的提取

实验前,在大量查阅各项相关研究资料的前提下,明确了CART与蛋鸡卵巢中不同大小黄白卵泡中的雌激素(Estrodiol, E2)和孕酮(P)分泌量的关系。本实验运用显微外科手术技术分离蛋鸡的大小黄卵泡、大小白卵泡,并分别进行总RNA的提取,通过CART mRNA表达量的差异对CART蛋白进行定位,再通过体外注射dsRNA对CART的表达进行抑制,阻断CART对蛋鸡卵巢中卵泡生长发育及排卵的影响,从而提高蛋鸡产蛋量,延长产蛋周期,降低料蛋比,进而通过饲养试验,确定调控效果,并将该技术进行全面示范推广。
According to the information of related research before experiments, we found out the relationships between the amount of estrogen(E2) and progesterone(P), which the layer’s follicle in different size secreted ,and CART. During this experiment ,we used the microsurgical technique to isolate different follicles including Rhubarb follicle(LYF) , Small white follicle (SWF), yellow follicle (SYF) and the follicle (LWF).Otherwise, we have picked up the total RNA respectively and located the CART protein through the different amount of CART expression. Finally, we have suppressed the influences of CART on the follicles’ growth and ovulation in ovarian by means of injecting dsRNA artificially .As to improve the layers’ production, prolong period of laying and reduce the ratio of forage and eggs, we conducted the feeding experiments and determined the effect of the results, and then apply comprehensively to the actual producatons.
